Early Expression and Supply (EES) - Part of Cell Culture & Fermentation Sciences (CCFS)


Description of work / project to be undertaken:

The Early Expression and Supply team is responsible for generating therapeutic proteins, usually antibodies, to support in-vitro and in-vivo studies at early project stages.

For the rapid production of these therapeutic proteins, AZ has developed a proprietary high-yielding, scalable and easy to use Chinese Hamster Ovary (CHO) cell based transient expression system.

This process is used at small scale (shake flasks) to assess expressibility of the proteins and at larger scales L rocking bioreactors) for material generation.

The team is constantly looking at ways to improve the process further focusing in the following areas:

optimisation of plasmid DNA production (in microbial systems), optimisation of the transfection conditions, assessment of impact of key process parameters, such us dissolved oxygen (DO) and pH on cell performance and protein yield, and finally on the robustness and scalability of the process in both rocking and stirred-tank reactors.

At the end of the internship the student will gain experience in microbial and CHO cell culture, plasmid DNA production, transient expression in bioreactors, experimental design, data analysis and in-depth understanding of biopharmaceutical development.


Primary duties of this position:


Gain experience in:


  • Attend and engage in department / team meetings, with opportunity to present
  • Plan and execute experiments, including relevant literature reviews. Analyse data and writeup experiments in electronic lab notebook.
  • Present findings, summarise outcomes and share future recommendations.
